PLCG1, or Phospholipase C Gamma 1, is a crucial enzyme protein involved in cellular signaling, differentiation, proliferation, and various other biological processes. it is crucial in receptor-mediated tyrosine kinase activator signaling within cells. Receptor tyrosine kinases interact with SRC domains to enzymatically activate PLCγ1 via phosphorylation at tyrosine 783. Upon activation PLCG1 leads to the translocation of Ras guanine nucleotide exchange factor RasGRP1 to the Golgi apparatus, activating Ras. PLCG1 is also a primary substrate of heparin-binding growth factor 1 (acidic fibroblast growth factor)-activated tyrosine kinases.

ICC analysis of Jurkat cells treated with pervanadate (50mM, 5mins) (top panel) and Jurkat cells untreated with pervanadate (50mM, 5mins) (below panel). Anti- Phospho-PLCγ1 (Tyr783) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
