A myc tag is a polypeptide protein tag derived from the c-myc gene product that can be added to a protein using recombinant DNA technology. It can be used for affinity chromatography, then used to separate recombinant, overexpressed protein from wild type protein expressed by the host organism. It can also be used in the isolation of protein complexes with multiple subunits.
Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized Histone H3, HA Tag and Myc tag transfected HeLa (Human cervix adenocarcinoma epithelial cell) labelling Myc tag (Alexa Fluor® 488 Conjugate) antibody at 1/2000 (0.1 μg) dilution/ (Right panel) compared with a Rabbit IgG Isotype Control / (Left panel). Then cells were stained with HA Tag - Alexa Fluor 647 antibody separately.
ICC shows positive staining in Histone H3-GST Tag-HA Tag-Myc tag transfected 293T cells. Anti-Myc tag (Alexa Fluor® 488 Conjugate) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
Negative control:ICC shows negative staining in vector-transfected 293T cells (below panel). Anti-Myc tag (Alexa Fluor® 488 Conjugate) antibody was used at 1/500 dilution and incubated overnight at 4°C. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
