A myc tag is a polypeptide protein tag derived from the c-myc gene product that can be added to a protein using recombinant DNA technology. It can be used for affinity chromatography, then used to separate recombinant, overexpressed protein from wild type protein expressed by the host organism. It can also be used in the isolation of protein complexes with multiple subunits.
Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized H3.3, Myc, His transfected 293T labelling Myc tag (Alexa Fluor® 647 Conjugate) antibody at 1/2000 (0.1 μg) dilution/ (right panel) compared with a Rabbit IgG Isotype Control / (left panel).
ICC shows positive staining in Myc-Histone H3.3 transfected HeLa cells. Anti-Myc tag (A674 conjugate) antibody was used at 1/200 dilution (magenta) and incubated overnight at 4°C. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
Negative control: ICC shows negative staining in vector-transfected HeLa cells. Anti-Myc tag (A674 conjugate) antibody was used at 1/200 dilution and incubated overnight at 4°C. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
