RIG-I (Retinoic Acid-Inducible Gene I) is a cytoplasmic RNA helicase that serves as a pattern recognition receptor in innate immunity. RIG-I can recognize pathogen-associated molecular patterns (PAMPs), particularly non-self viral RNA, activating the RIG-I signaling pathway to promote cytokine production and exert antiviral effects. RIG-I primarily detects double-stranded 5'-triphosphate RNA (3pRNA) during viral or bacterial infections. Once bound to ligands in myeloid cells, RIG-I recruits an adaptor protein called mitochondrial antiviral signaling protein (MAVS) to induce the production of pro-inflammatory cytokines and type I interferons (IFN-I), as well as ASC-dependent inflammasome activation, leading to a variety of innate and adaptive immune responses. RIG-I activation is crucial for the efficacy of immune checkpoint blockade in anti-tumor therapy. Activation of the RIG-I pathway in myeloid cells can induce IFN-I-dependent natural killer cell activation and regression of metastases derived from primary melanoma.

ICC shows positive staining in RAW264.7 cells. Anti- Rig-I/DDX58 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
