JAK3, which stands for Janus Kinase 3, is a non-receptor tyrosine kinase that plays a crucial role in the immune system. It is predominantly expressed in immune cells such as T cells, B cells, and natural killer cells. JAK3 is involved in the intracellular signaling pathways initiated by cytokine receptors, particularly those of the common gamma chain family, including interleukin-2 (IL-2), IL-4, IL-7, IL-9, IL-15, and IL-21 receptors. Upon receptor activation, JAK3 phosphorylates downstream signaling molecules, leading to the activation of transcription factors like STATs and ultimately regulating cell proliferation, differentiation, and survival. Mutations in the JAK3 gene can result in severe combined immunodeficiency (SCID), highlighting its essential function in immune development and function. Additionally, due to its central role in immune signaling, JAK3 has become a target for therapeutic interventions in autoimmune and inflammatory diseases.
