HLA-DR is a subtype of the human leukocyte antigen (HLA) complex, specifically classified as a class II molecule, and is encoded by genes located on chromosome 6. It plays a crucial role in the immune system by presenting peptide antigens to CD4+ T cells, which subsequently trigger adaptive immune responses, including the activation of B cells and the production of antibodies. HLA-DR is primarily expressed on antigen-presenting cells such as macrophages, dendritic cells, and B cells, and its expression levels can indicate the degree of immune system activation. This molecule is also significant in clinical settings, as it is associated with various autoimmune diseases (e.g., rheumatoid arthritis linked to HLA-DR4) and is a key factor in determining the success of organ transplants. Additionally, HLA-DR has been studied in the context of cancer immunotherapy, where its expression on tumor cells can influence the effectiveness of immune-mediated tumor suppression.

IHC shows positive staining in paraffin-embedded human liver. Anti-HLA-DRA antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
IHC shows positive staining in paraffin-embedded human tonsil.
IHC shows positive staining in paraffin-embedded human colon.
IHC shows positive staining in paraffin-embedded human lung cancer. Anti-HLA-DRA antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). ICC shows positive staining in Raji cells (top panel) and negative staining in 293T cells (below panel). Anti- HLA-DRA antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
