Collagen I protein is the most abundant collagen in the human body, making up around 90% of the total collagen in vertebrates. It is a major structural component of connective tissues, including skin, tendons, ligaments, bones, and the organic part of teeth. Collagen I is composed of a triple helix structure formed by two α1 chains and one α2 chain encoded by the COL1A1 and COL1A2 genes, respectively. This protein provides significant tensile strength and elasticity to tissues, allowing them to withstand mechanical stress. Its synthesis involves complex processes, including transcription, translation, post-translational modifications, and assembly into collagen fibers. Due to its high biocompatibility and unique physical properties, Collagen I is widely used in pharmaceuticals, regenerative medicine, and clinical applications.

IHC shows positive staining in paraffin-embedded human kidney. Anti-Collagen I antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ICC shows positive staining in HFF cells (top panel) and negative staining in HT-29 cells (below panel). Anti-Collagen I antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
