MG53, also known as TRIM72, is a member of the TRIM protein family, primarily expressed in the heart and skeletal muscle. It consists of an N-terminal RING, B-box, and curly helix region, with a C-terminal SPRY domain. MG53 plays a crucial role in acute cell membrane repair, anti-inflammation, stem cell regeneration, and anti-virus effects. In acute cell membrane injury, MG53 oligomerizes and recruits intracellular vesicles to the injury site, forming membrane repair patches. It also inhibits the NF-κB pathway to reduce inflammation and promotes stem cell survival and regeneration. MG53 has shown therapeutic potential in myocardial infarction, acute lung injury, and other tissue injuries.
