Eukaryotic elongation factor 2 (eEF2) is a crucial regulator of protein synthesis, belonging to the highly conserved guanosine triphosphatase (GTPase) family. It is essential for the elongation phase of protein translation, facilitating the GTP-dependent translocation of the nascent protein chain from the A-site to the P-site of the ribosome. eEF2's activity is tightly regulated by phosphorylation at Thr56 by eEF2 kinase (eEF2K), which inhibits its function and thus suppresses protein synthesis. This protein is ubiquitously expressed in various tissues and plays a significant role in normal cellular functions. However, its dysregulation is implicated in several diseases, including cancer, where overexpression of eEF2 is associated with increased tumor incidence and poorer patient prognosis.
