  • MICB (MHC class I polypeptide-related sequence B) is a stress-inducible, heavily glycosylated transmembrane protein encoded by the MICB gene located on chromosome 6p21.33. It functions as a ligand for the NKG2D receptor, which is expressed on natural killer (NK) cells, CD8+ T cells, and γδ T cells. Upon binding, MICB activates the cytolytic response of these immune cells, promoting their ability to target and eliminate stressed or transformed cells, such as tumor cells or virus-infected cells. Unlike classical MHC class I molecules, MICB does not associate with β-2-microglobulin or bind peptides. Its expression is induced by cellular stress, including DNA damage and viral infections, making it a crucial component in both innate and adaptive immune responses. MICB is widely expressed in various tissues, including lymph nodes and bone marrow, highlighting its role in diverse physiological contexts.

    • ICC shows positive staining in K562 cells (top panel) and negative staining in THP-1 cells (below panel). Anti-MICB antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
