NDP52, also known as CALCOCO2, is a key autophagy receptor protein involved in selective autophagy processes, playing a crucial role in the recognition and degradation of invading pathogens and damaged organelles such as mitochondria. It acts as a bridge linking target substrates to autophagosomes through interactions with ubiquitinated cargos and LC3, a component of the autophagosome membrane. NDP52 is also implicated in immune signaling, where it can regulate Toll-like receptor (TLR) pathways and has been shown to mediate antiviral responses by targeting viral components for lysosomal degradation. Additionally, NDP52 has been found to interact with various proteins, including TBK1, NAP1, and Myosin VI, highlighting its multifaceted role in cellular homeostasis and defense mechanisms.
