CD47,也被称作整合素相关蛋白(Integrin-associated protein, IAP),是所有类型细胞表达的靶点中“marker of self”的典型代表。CD47是一个糖蛋白,具有免疫球蛋白可变的N端结构域、5个跨膜结构域和一个短的C端胞内尾,胞内尾部有四种可变不同剪接异构体,从而形成4个亚型。CD47是靶向肿瘤免疫治疗中发挥主要作用的免疫细胞类群,癌细胞表达的CD47与NK细胞表达的SIRP家族蛋白SIRPα以及SIRPγ相互作用,通过这样的方式保护癌细胞避免被吞噬消除。CD47在多种实体瘤细胞及恶性血液瘤细胞上呈现高表达,而且其表达水平与疾病进展呈正相关。
