Angiopoietin-like protein 3 (ANGPTL3) is a multifunctional secreted factor predominantly expressed in the liver and subject to various post-translational modifications, including cleavage and glycosylation. It comprises a signal peptide, an N-terminal coiled-coil domain, and a C-terminal fibrinogen-like domain. While the N-terminal domain regulates lipid metabolism, the C-terminal domain is implicated in angiogenesis. ANGPTL3 is crucial for biological processes such as lipid homeostasis, blood vessel formation, and hematopoietic functions. It is also associated with pathological conditions like atherosclerosis, cancer, nephrotic syndrome, diabetes, and liver diseases, making it a potential biomarker. ANGPTL3 signaling pathways, involving LXR, thyroid hormone, insulin, and leptin, contribute to both physiological and pathological processes.
