背景介绍
  • CD44 is a widely expressed transmembrane glycoprotein on the cell surface, belonging to the adhesion molecule family. It primarily functions as a receptor for hyaluronic acid (HA) and mediates cell-cell and cell-matrix interactions. Its structure includes variant exon-spliced isoforms (e.g., CD44v) and a constant region (CD44s), with post-translational modifications such as glycosylation generating multiple variants. CD44 plays crucial roles in physiological and pathological processes, including inflammation, wound healing, lymphocyte homing, and tumor metastasis. It is highly expressed in cancer stem cells (CSCs) and is associated with tumor progression, drug resistance, and poor prognosis. Key signaling pathways involving CD44 include PI3K/AKT and RAS/MAPK, making it a potential therapeutic target in cancer and stem cell research.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (red).

    • ICC shows positive staining in PANC-1 cells. Anti-CD44 antibody was used at 1/250 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).

    • Negative control: ICC shows negative staining in LNCaP cells. Anti-CD44 antibody was used at 1/250 dilution and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
