P53 is a gene that encodes for the p53 tumor suppressor protein, commonly referred to as the “Guardian of the Genome” [PMID: 1614522]. The p53 protein is a transcription factor that controls the output of many biological processes according to the type of cellular stress signal input. Stress signals known to activate p53 include oncogene activation, DNA damage, and replication stress [PMID: 28886379]. In response to these stresses, p53 undergoes post-translational modifications, promotes the transcription of genes involved in specific cell responses according to the stress type thereby controlling the cell’s fate [PMID: 15510160]. When the p53 protein is mutated or aggregated, it loses its function, resulting in tumor progression and growth [PMID: 29789497].
IF shows positive staining in paraffin-embedded human colon cancer. Anti-p53 (Alexa Fluor® 555 Conjugate) antibody was used at 1/100 dilution (Red) and incubated overnight at 4°C. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
