TMEM119 is a cell surface protein that serves as a specific marker for microglial cells in both mice and humans. It functions as an osteoblastic inductive factor, promoting the proliferation, migration, and invasion of osteosarcoma cells. TMEM119 also acts downstream of parathyroid hormone (PTH) and is similar to the similar to mothers against decapentaplegic-3 (Smad3) signaling pathway in osteoblasts. TMEM119 is also considered a potential microglial cell marker that can distinguish resident microglia from blood-derived macrophages in the human brain. It is crucial for identifying microglia in healthy tissue and distinguishing them from infiltrating macrophages and other cell types in neurodegenerative disease models.
Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized SH-SY5Y (Human neuroblastoma epithelial cell) labelling TMEM119 antibody at 1/500 dilution (0.1 μg)/ (Red) compared with a Rabbit monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-TMEM119 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
IHC shows positive staining in paraffin-embedded human tonsil.
IHC shows positive staining in paraffin-embedded human testis.
Negative control: IHC shows negative staining in paraffin-embedded human liver.
IHC shows positive staining in paraffin-embedded human colon cancer.
ICC shows positive staining in SH-SY5Y cells. Anti-TMEM119 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
