The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with
fluorecence-labeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were
separated and detected by MSA device. The conversion rate of ERK1 is 30% when it reacts with 20 μM ATP
and 2 μM Peptide 8 for 30 minutes, and the corresponding enzyme concentration is 25 nM.
