The interleukin-6 receptor (IL-6R) comprises an 80-kDa IL-6-binding protein (α chain/CD126) and gp130. CD126, a glycoprotein with the ligand-binding site, is part of the human IL-6R, which spans 468 amino acids, including a signal peptide, extracellular region, transmembrane domain, and a short cytoplasmic domain of 19, 339, 28, and 82 amino acids, respectively. IL-6R is present on multiple myeloma, hepatocellular carcinoma, prostate carcinoma, and epidermoid carcinoma cells. The IL-6/IL-6R pathway drives tumor growth, particularly in gliomas, making IL-6R a drug delivery target and a therapeutic target. Primary human cartilage cells express IL-6R, which is upregulated by IL-6, positioning cartilage cells as direct targets of IL-6.
